Prolific: What's Under the Hood

Here's a closer look at each:
Citrulline
Citrulline is an amino acid and precursor to another amino acid called arginine.
Arginine is commonly included in pre-workouts since it directly produces nitric oxide, an important molecule that relaxes the inner muscles of the blood vessels, causing them to widen and increase circulation.
However, arginine is subject to first-pass metabolism by the intestines and liver, meaning a large percentage of it is broken down before it gets a chance to reach the bloodstream.
On the other hand, citrulline is not subject to this same fate and actually inhibits the enzyme that breaks down arginine.
For this reason, citrulline supplementation increases arginine levels in the body more than supplementing with arginine itself and is therefore much more effective for increasing nitric oxide (1).
To this point, dozens of clinical studies have demonstrated that citrulline supplementation at or below the dose provided in Prolific can delay muscle fatigue and increase endurance during high-intensity strength training, with other studies demonstrating improved post-exercise muscle recovery (2, 3, 4).
Betaine
Betaine — or trimethylglycine (TMG) — is an anti-inflammatory and antioxidant compound.
It was first isolated from beets Beta vulgaris, from which it's named.
Betaine sustains cell hydration and function in response to various physiological stressors — including exercise — increases free choline thereby potentially increasing acetylcholine production to support muscle contractions, and may influence certain additional mechanisms leading to downstream muscle growth (5, 6, 7).
Through these actions, studies have shown that supplementation with 2.5 grams of betaine — the dose provided in Prolific — daily for around two weeks significantly improves muscle strength, reduces muscle fatigue, and improves body composition (8, 9, 10, 11).
Taurine
Taurine is an amino acid heavily concentrated in the brain, heart, and skeletal muscles.
Despite the common belief, taurine does not come from bull semen or urine, but it was first isolated from the bile of an ox. Bos taurus, the Latin name for an ox, is where its name originated.
By maintaining proper hydration and electrolyte balance in cells, regulating levels of minerals necessary for muscle contractions within your cells like calcium, and through its antioxidant function, taurine supplementation may improve exercise performance and decrease exercise-induced muscle damage and oxidative stress (12, 13, 14, 15).
Other research suggests that taurine supplementation can improve lipid metabolism with exercise by affecting genes related to mitochondrial activity and fatty acid oxidation as well as improve thermoregulatory capacity during exercise in hot conditions (16, 17).
These benefits have been demonstrated using the same dose found in Prolific.
Caffeine
Caffeine is the star of nearly all pre-workouts — but for good reasons.
Dozens of clinical studies have demonstrated that caffeine ingestion significantly enhances muscle endurance, strength, and power in several ways, including intracellular calcium release in muscle cells to increase muscle force, preservation of muscle glycogen, and antagonistic action to adenosine's depressant effects in the brain (18).
Caffeine can also improve mood and measures of cognitive functions like attention when consumed before exercise (19).
Caffeine tends to be most beneficial for improving exercise performance when consumed at a dose of three to six milligrams per kilogram of body weight (20).
This translates to 200 to 400 mg of caffeine for a 150-pound (68-kg) person.

Theanine is an amino acid found primarily in green and black tea and certain types of mushrooms.

Citicoline is a precursor to phosphatidylcholine, a major cell membrane phospholipid that supports the production of adenosine triphosphate (ATP) or energy in the brain to maintain focus.

While "bro-sciencey" sounding, actively focusing on the muscle squeeze and contraction during lifts has been shown to increase muscle fiber recruitment and activation, supporting greater gains in muscle and strength (25, 26, 27).
Rhodiola
Rhodiola (Rhodiola rosea) is an herb that has a long history of use in traditional medicine for increasing endurance and work performance, and to manage fatigue, weakness, and other symptoms (28).
It's categorized as an adaptogen, meaning it increases the body's resilience to stress in nonspecific ways.
Through its adaptogenic properties, rhodiola supplementation has been shown to improve antioxidant activity and reduce muscle fatigue and post-exercise muscle soreness and damage (29, 30).
Coffee fruit extract
Coffee fruit extract, as the name suggests, comes from the fruit of the coffee plant.
